摘要
目的:用荧光偏振免疫分析法测定白血病患者血清中甲氨蝶呤的浓度。方法:对5例白血病患者静滴大剂量甲氨蝶呤(1000mg)的药物动力学进行研究。结果:分布半衰期(t1/2α)为(1.63±0.23)h,β相消除半衰期(t1/2β)为(12.8±2.4)h,速率常数(K10)为(0.41±0.05)/h,表观分布容积(Vc)为(17±5)L,总清除率(Cls)为(6.8±1.3)L/h,药时曲线下面积(AUC)为(334±61)μmol·h/L,其经时血药浓度符合二室开放模型。结论:结果可靠,与文献报道一致,可用于临床用药参考。
Objective: To measure the serum concentration of Methotrexate with fluorescence polarization immunoassay. Methods: The pharmacokinetics of five leucocythemia patients received high-dose Methotrexate (1 000 mg) were studied. Results: t1/2α was (1.63±0.23) h, t1/2β was (12.8±2.4) h, K10 was (0.41±0.05)/h, Vc was (17±5) L, Cls was (6.8±1.3) L/h, AUC was (334±61)μmol·h/L, and its blood concentration was consistent with the two-compartment open model. Conclusion: The results is credible and consistent with reported literatures, and can be used as a reference in clinic.
