摘要
目的:观察黄芪多糖(APS)联合顺铂(DDP)对小鼠H22肝癌移植瘤生长的抑制作用。方法:复制H22肝癌小鼠移植瘤模型,将40只接种H22肝癌细胞昆明小鼠随机分成4组,其腹腔分别注射生理盐水(对照组)、ASP组、DDP组以及APS+DDP组,观察小鼠的毒副反应以及生存质量。实验19d后,处死全部小鼠,剥离皮下肿瘤,称小鼠肿瘤重量,计算出抑瘤率。结果:APS单用组的H22肝癌平均瘤重为(1.29±0.29)g,明显低于对照组的(1.86±0.73)g(t=3.9746,P<0.001)。APS+DDP组的H22肝癌平均瘤重为(0.52±0.31)g,均明显低于APS单用组(t=9.9351,P<0.001)和DDP单用组(t=4.1970,P<0.001);其抑瘤率达72.0%,明显高于APS单用组(χ2=17.3756,P<0.001)和DDP单用组(χ2=5.0794,P<0.05)。APS+DDP组小鼠毒副反应明显低于DDP组,生存质量优于DDP组。结论:黄芪多糖和顺铂合用对小鼠H22肝癌移植瘤的生长具有协同抑制作用,并能降低顺铂毒副反应,提高生存质量。
Objective:To research the inhibition action induced by Astragalus Polysaccharides(APS) with cisplatin(DDP) in treating H22 liver cancer in mice. Methods:H22 liver cancer cells were inoculated into the mammary fatty pad of mice to establish H22 liver cancer model,40 mice bearing highly metastatic H22 liver cancer cells were randomized into four groups:control group,DDP group,APS group,APS+DDP group. Different treatments were served after 7 days transplantation and all mice were sacrificed after 19 days. During the treatments,the toxic side reaction of chemotherapy and the life quality were observed. Subcutaneous tumors were processed for tumor weight,and tumor growth inhibition rate and tumor necrosis rate of each group were calculated.Result:The tumor weight in APS group was significantly lower than that in contool group [(1.29±0.29)g vs. (1.86±0.73)g,t=3.974 6,P〈0.001].The tumor weight in APS+DDP group [(0.52±0.31)g] was significantly lower than that in APS group or DDP group(t=9.935 1,P〈0.001)and (t=4.197 0,P〈0.001).The tumor inhibition rate in APS+DDP group (72.0%) was significantly higher than that in APS troup or DDP group(χ^2=17.375 6,P〈0.001)and (χ^2=5.079 4,P〈0.05). Compared with DDP group,the toxic side reaction of chemotherapy in combination group was significantly lower,but life quality higher.Conclusion:Combined application of APS and DDP has a synergic anticancer effect on H22 liver cancer in mice,and can decrease toxic side reaction of chemotherapy and improve life quality.
