大鼠骨髓源性肝干细胞形态学特征的实验研究 被引量:1

摘要 目的:探讨大鼠骨髓源性肝干细胞的形态特性和来源。方法:HGF体外诱导大鼠骨髓干细胞向肝样细胞分化,诱导第0、7、14、21天,相差显微镜观察细胞形态特征,电镜观察第21天诱导组与非诱导组细胞超微结构变化;免疫细胞化学法检测甲胎蛋白(AFP)、细胞角质蛋白18(CK18)的表达;PAS检测细胞糖原。结果:HE染色较深的小圆形细胞在非诱导条件下各时相直径未发生明显变化,AFP、CK18、糖原染色均为阴性;诱导组HE染色较深的小圆形细胞与AFP、CK18、糖原染色阳性的细胞形态大小一致,随诱导时间延长其直径增大。透射显微镜观察:第21天诱导组细胞间有大量的微绒毛,细胞内可见到丰富的粗面内质网、线粒体、溶酶体、吞饮泡;第21天非诱导组细胞间仅有少量微绒毛,细胞质较少。结论:骨髓源性肝干细胞很可能来源于集落中HE染色较深的小圆形细胞,在HGF诱导下可以分化为肝样细胞,其形态学及超微结构与肝细胞结构相符合。
