摘要 为评价荔枝种质的遗传多样性,从88对SRAP引物中筛选出22对多态性高、稳定性好的引物,用于研究46个荔枝品种。每对引物组合产生6~19条谱带,其中多态性条带284条,占总带数的97.93%。46个种质的相似系数在0.469~0.838之间,平均相似系数为0.644。其中,秤砣与红荔的相似性系数最大,亲缘关系最近;小丁香与三月红、三月红与A13两对相似系数最小,亲缘关系最远。UPGMA聚类分析显示,在相似系数为0.644处46份荔枝供试品种被划分为4个类群,并且来源地相同的品种间亲缘关系较密切。22对多态性引物中,引物me7-em2在海垦18中扩增出1条特征谱带,引物me3-em7在水东中也扩增出1条特征谱带,而仅凭这1条特征带就可以将荔枝品种海垦18或水东与其它材料区别开来。这说明SRAP标记可用于荔枝的品种鉴定和群体遗传结构的研究。 To evaluate the genetic diversity of litchi, 46 accessions of litchi were analyzed by 22 highly polymorphic and stable primer pairs selected from 88 pairs of SRAP primer. With each primer combination generated 6-19 bands, 284 polymorphic bands were detected, accounting for 97.93% of the total. The similarity coefficient of the 46 litchi germplasm was 0.469-0.838, with the average similarity coefficient of 0.644. In all varieties, the Pengtuo and the Hongli had the greatest similarity efficient. Two pairs of litchi (Xiaodingxiang and Sanyue- hong, Sanyuehong and A13) had the minimum similarity coefficient. 46 litchi germplasms were divided into four groups on the similarity level of 0.644. Varieties of the same origin had a closer relationship. Of the 22 polymorphic primers combinations, both me7-em2 and me3-em7 had specific amplification bands in Haikenl 8 and Shuidong, respectively, which could distinguish them from others. It is demonstrated that SRAP markers can be used for variety identification and genetic construction of litchi.
