摘要
目的从CM10、IMAC3-Cu这两种芯片中选择适合应用于血清蛋白质指纹图谱研究的芯片种类。方法选择慢性乙型肝炎患者、肝硬化患者和健康人血清各13例,采用CM10、IMAC3-Cu两种蛋白芯片,经SELDI-TOF-MS技术对样本进行检测比较。结果在2000-50000Da范围内,CM10和I-MAC3-Cu芯片在各实验组捕获的有效蛋白峰数的差异均有统计学意义(P〈0.05)。结论CM10能得到更多慢性乙型肝炎患者、肝硬化患者和健康人的有意义的蛋白峰值,能更好的应用于血清蛋白组学的实验研究。
Objective To select a suitable protein chip for detection of serum protein fingerprint from two kinds of chips(CM10 and IMAC3-Cu).Methods Serum samples from 13 patients with chronic hepatitis B,13 patients with liver cirrhosis and 13 normal control people were collected respectively,peptide spectra were determined by SELDI-TOF-MS measurement.Results In the range from 2000Da to 50000Da,the number of significant protein peaks captured in each experimental group has a significant difference between CM10 and IMAC3-Cu chips(P〈0.05).Conclusion CM10 chips can detect more significant difference protein peaks compared with IMAC3-Cu chips and preferably apply to the research of the serum protein fingerprints.
