摘要
由于hG-GF的重要作用以及rhG-CSF在临床上的广泛使用,有必要进一步了解正常人和某些疾病状态下病人外周血循环中G—CSF的浓度。利用两株针对不同抗原决定簇的单克隆抗体,建立了双抗夹心ELISA方法。方法特异性较好,检测灵敏度为340pg/ml。用该法检测了12例细菌感染病人,有11例病人G—GF检测阳性,阳性率91.6%;白血病病人血清G—GF阳性率25%~35.7%,正常血清则均为阴性。该方法克服了以往骨髓细胞集落形成法和G—GF依赖株测定法需时长、操作繁琐的缺点,为研究正常人和某些疾病状态下病人外周血G—CSF浓度的变化和G—CSF的药代动力学提供了一个简便、快速、特异性高的方法。
The double antibody sandwich ELISA was established by using these McAbs for detecting G-CSFlevels in human serum. The assay's sensitivity was 340 pg/ml. G-CSF levels of human serum samples were alsodetected, it was found that the positive rates of G-CSF were zero in normal serum samples and 91. 6 % in bac-terial infection patients and 25% ~ 35. 7 % in leukemia patients. The research provided a simple, rapid, specifici-ty method for detecting the level of human serum G-CSF. The method overcame the flaw of myeloid progenitorcells assay and NFS- 60 cells assay which were tedious and time-consuming.
