摘要
目的:探讨异搏定、潘生丁、丹参、黄体酮、环孢菌素A和他莫西芬等药物对耐阿霉素人卵巢癌细胞系A2780/ADM逆转的耐药性。方法:用MTT法和高效液相色谱仪测定异搏定等6种药物分别对阿霉素耐药细胞的敏感性和耐药细胞内阿霉素的剂量。结果:单用异搏定等6种药物对A2780/ADM几乎无体外抑制作用。异搏定和环孢菌素A对A2780/ADM有明显的增效作用,可增加ADM在A2780/ADM细胞内的潴留浓度。结论:除丹参外,其它5种药物均能增强ADM对A2780/ADM耐药细胞细胞毒作用,并克服ADM的耐药性。
Objective:To investigate the reversional action to ADM resistance in isoptin,persantin,Dansan,progesteronum,cyclosporin A and tamoxifen with A2780/ADM human ovarian cancer cell lines which have resistance to ADM.Methods:MTT method and high performance liquid chromatographic assay(HPLCA)were utilized to analyse the concentration of ADM in A2780/ADM cells and these cells' sensitivity to ADM when adding isoptin,and so on(totally six drugs)respectively.Results:No inhibitory effect to A2780/ADM cell were found in isoptin and so on six drugs.Isoptin and cyclosporin A increased the concentration and the sensitivity of A2780/ADM to ADM.Conclusions:Five drugs(except Dansan)could increase the inhibition and decrease the resistance of A2780/ADM to ADM.It is possible to use these five drugs to increase the effects of ADM in clinical treatment of patients with ovarian cancer.
