延肾口服液对人肾小球系膜细胞增殖及分泌FN、LN、Ⅳ-c的影响 被引量:1

摘要 【目的】观察中药复方制剂延肾口服液(YSH)对培养的人肾小球系膜细胞(huMsC)增殖及分泌细胞外基质(ECM)的影响,探讨其防治肾小球硬化的作用机制。【方法】采用四甲基偶氮唑盐法(MTT)比色法和双抗夹心ELISA法,检测灌服延肾大鼠不同浓度药物血清对培养的人肾小球系膜细胞的增殖及分泌纤维连接蛋白(FN)、层连蛋白(LN)和Ⅳ型胶原(Ⅳ-c)的影响。【结果】5%-20%YSH大鼠血清对原代培养的人肾小球系膜细胞增殖及产生FN、LN、Ⅳ-c有明显的抑制作用(P〈O.05或P〈0.01),其抑制程度与药物浓度有-定的量效关系。【结论】延肾制剂对培养的人肾小球系膜细胞增殖及产生FN、LN、Ⅳ-C的抑制作用,是其防治肾小球硬化发生发展的重要作用机制之-。 [Objective] To observe the effect of traditional chinese drugs- Yanshen (YSH) on prohferation of mesangial cells (MsC) and production of the extracellular matrix produced by MsC cultured in vitro. [Methods] The effects of the different concentration of YSH rat serum on proliferation of MsC and the production of the fibronectin (FN), laminin (LN) and Ⅳ- collagen (Ⅳ- c) which is produced by MsC cultured in vitro were observed. MTT colourmetry and ELISA method were used. [Results] Prohferation of MsC and the production of FN, LN and Ⅳ-c can be significantly inhibited by 5% - 20% YSH rat serum ( P 〈 0.05 or P 〈 0.01 ) and inhibition rate is relative to drug concentration. [Conclusions] It is one of the mechanism on preventing the occurence and development of glomerulosclerosis that prohferation of MsC and the production of FN, LN and Ⅳ-c which produced by MsC were inhibited by YSH.
