二氢埃托啡自动静脉给药对大鼠免疫功能的影响

EFFECT OF DIHYDROETORPHINE ON IMMUNE FUNCTIONS IN RATS TREATED BY AUTOMATICAL INTRAVENOUS INJECTION

摘要 本研究采用计算机实时控制的定时自动静脉给药法,72h快速致使大鼠身体依赖性形成。各组戒断症状分值为:NS组11.0±s2.9;吗啡组81.0±s8.6;DHE组41.0±s4.4,表明大鼠已形成明显身体依赖性(P<0.001)。在该模型基础上,利用3H-TdR掺入法测得DHE明显抑制大鼠淋巴细胞增殖反应。各组刺激指数为:80.69±s1.76;49.04±s1.10和30.77±s2.14(P<0.001)。利用MTT法测DHE也能抑制大鼠IL-2活性,各组A570nm值为:0.39±s0.03;0.31±s0.04和0.26±s0.04(P<0.01)。结果表明DHE自动静脉给药明显抑制大鼠免疫功能。 Computer controlled automatical intravenous injection was adopted to establish physical dependence in DHE treated rats. The withdrawal scores of 2 opiate-dependent rat groups were: DHE group 41.0±s 4.4; Morphine group 81.0±s 8.6. Both showed significant differences compared with that of NS group (11.0±s 2.9, P< 0.001).Based on this modal, effects of DHE on immune functions were examined. DHE significantly suppresses the Con-A activated splenic lymphocyte proliferations in rats. The stimulation indexes were: DHE group 30.77±s 2.14; Morphine group 49.04±s 1.10.Both showed significant differences compared with that of NS group (80.67±s 1.76, P< 0.001).DHE can also suppress the activity of IL-2 in the supernatant of Con-A activated splenocytes in rats. The values of A570nm were: DHE group 0.26±s 0.04; Morphine group 0.31±s 0.04. Both showed differences compared with that of NS group (0.39±s 0.03, P<0.01).The results indicate that DHE has powerful immunosuppressive effects in rats; furthermore, its effects are at least as 1000 times as those of morphine at the same dose.
