硫辛酸和维生素E对氢醌所致V79细胞毒性及线粒体膜电位影响的干预性实验

摘要 目的研究氢醌对V79细胞的毒性作用和线粒体膜电位的影响,探讨硫辛酸和维生素E的干预性作用。方法采用MTT和荧光探针JC-1,分别测定一定浓度氢醌与硫辛酸和维生素E单独或联合作用对细胞存活率及线粒体膜电位的影响。结果与对照组比较,50μmol/L和100μmol/L氢醌可导致V79细胞存活率和线粒体膜电位的下降;与单纯氢醌组比较,硫辛酸和维生素E能明显拮抗50μmol/L和100μmol/L氢醌所引起的V79细胞存活率的下降,并且50μmol/L的硫辛酸可以提高50μmol/L氢醌所引起的V79细胞线粒体膜电位的下降,而25μmol/L的维生素E对50μmol/L氢醌引起的V79细胞线粒体膜电位的下降没有影响。结论氢醌对V79细胞具有细胞毒性和线粒体毒性作用;一定浓度的硫辛酸和维生素E都可以拮抗氢醌对V79细胞产生的毒性作用。提示,硫辛酸可能具有线粒体靶向保护作用。 Objective To explore the cytotoxicity effects of hydroquinone on V79 cells and the intervention effects of lipoic acid and vitamin E. Methods The viability and the levels of mitochondrial transmembrane potential( △ψm) of V79 cells were analyzed by the MTT assay and JC-1 assay. Results Compared with the control group, 50 and 100 μmol/L hydroquinone could decrease cell viability and △ψm significantly, while lipoic acid or Vitamin E could effectively antagonize V79 cells from cell toxicity induced by hydroquinone when compared with the hydroquinone group. 50 μmol/L lipoic acid could prevent from △ψm dropping induced by 50 μmol/L hydroquinone; however, 25 μmol/L vitamin E didn' t show this effect. Conclusions Hydroquinone showed a cytotoxicity and mitochondrial toxicant in V79 cell, a certain level of lipoic acid and vitamin E could antagonize cytotoxic effect on V79 cell induced by hydroquinone. The data suggest that lipoic acid might have target protection effect on △ψm induced by hydroquinone.
