Simultaneous Determination 5 Contents of the Rhubarb in the Liaojin Film by HPLC

摘要 目的建立高效液相色谱法同时测定疗筋涂膜剂中5种成分的含量。方法采用Hypersil ODS-C18柱(250mm×4.6mm,5μm),甲醇-0.05%磷酸(85∶15)为流动相,流速:0.8mL·min-1,柱温:34℃,检测波长:254nm。结果芦荟大黄素在9.11~145.76μg·mL-1(r=0.9994),大黄酸在5.63~90.08μg·mL-1(r=0.9993),大黄素在5.44~87.04μg·mL-1(r=0.9999),大黄酚在6.13~98.08μg·mL-1(r=0.9999),大黄素甲醚在2.72~43.52μg·mL-1(r=0.9998)线性良好;平均回收率分别为97.64%、98.14%、99.52%、99.36%、98.43%;RSD分别为1.29%、1.81%、1.40%、0.59%、1.58%。结论该方法简便、可靠、准确,可用于疗筋涂膜剂的质量控制。 OBJECTIVE To establish an HPLC method for the determination of Chrysophanol, Emodin, Physcion, Aloeemodin and in Liaojin film. METHODS The determination was performed on Hypersil ODS-C18 column(250 mm × 4.6 mm, 5 μm)with mobile phase consisted of methanol-0. 05 % phosphoric acid (85 : 15)at a flow rate of 0.8 mL·min^-1. The UV detection wavelength was set at 254 nm. RESULTS The calibration curves were linear in the range of 9.11-145.76 μg·mL^-1 (r=0.999 4) for Aloeemodin, 5.63-90.08 μg·mL^-1 (r=-0.999 3 ) for Rhein, of 5.44-87.04 μg·mL^-1 (r=-0.999 9) for Emodin, 6.13-98.08 μg·mL^-1 (r=-0.999 9)for Chrysophanol and 2.72-43.52 μg ·mL^-1(r=-0.999 8) for Physcion. The average recovery was 97.64%, 98.14%, 99.52%, 99.36%, 98.43%; RSD wasl.29%, 1.81%, 1.40%, 0.59%, 1.58%.CONCLUSION The determination method is simple, reliable, accurate, and it can be used for quality control of Liaojin film.
