摘要
目的探讨凋亡相关基因在骨肉瘤细胞中的表达及其临床意义。方法采用3个公认的骨肉瘤细胞株及1个成骨细胞株,提取总RNA,合成生物素标记的cRNA与Affymetrix Ge—neChip U133A芯片杂交,筛选差异表达的凋亡相关基因。利用MATLAB软件进行细胞凋亡通路的KEGG分析。结果以表达差异≥2.0倍为限,在Affymetrix HG—U133A芯片包含的所有基因中,3个骨肉瘤细胞株与成骨细胞株比较,一共筛选出7个差异表达的凋亡相关基因:3个上调基因(IKBKB、IL1R1和FAS),4个下调基因(TP53、PPP3CB、PPP3CC和APAF1)。利用MATLAB软件将7个差异表达的凋亡相关基因映射到KEGG的凋亡通路上。结论骨肉瘤是细胞增殖和细胞凋亡间平衡失调的结果,通过调控细胞凋亡相关基因的表达可以诱导肿瘤细胞凋亡,达到抗肿瘤目的。
Objective To explore the expression of apoptosis related genes in osteosareoma cells, and study their clinical significance in osteosarcoma. Methods Three well-known ATCC osteosarcoma cell lines and an osteoblastic cell line were collected. After the total RNA extraction,the finally synthesized biotinylated cRNAs were hybridized to Affymetrix GeneChip U133A arrays. Bioinformatics method was used to screen the apoptosis related genes which were differentially expressed. Results Using an entrance limit of ≥ 2.0,there were 3 up-regulated (IKBKB, ILl R1 and FAS) and 4 dowm-regulated apoptosis related genes (TP53 ,PPP3CB,PPP3CC and APAF1 ) identified by microarray which were commonly differentially expressed among all the three osteosarcoma cell lines in comparison with the osteoblastic cell line hfob1. 19. Conclusion Osteosarcoma is a result of disequilibrium between cell proliferation and apoptosis. Regulating the expression of apoptosis related genes may induce apoptosis of tumor cells, and achieve the purpose of anti-tumor.
