利金方对慢性阻塞性肺疾病肺气虚证大鼠IL-8及TNF-α表达的影响  被引量:8

Effects of Liking Prescription on IL-8,TNF-α Expression of Chronic Obstructive Pulmonary Disease Qi Deficiency Syndrome Rats

摘要 目的观察利金方对慢性阻塞性肺疾病(COPD)肺气虚证大鼠血清和支气管肺泡灌洗液(BALF)中白细胞介素-8(IL-8)、肿瘤坏死因子-α(TNF-α)表达的影响。方法将50只SPF级W istar大鼠随机分为空白组、模型组、补肺汤组、利金方高剂量组及利金方低剂量组,每组10只。采用复合因素造模法建立COPD大鼠模型。各药物组分别以相应剂量连续给药14d,检测各组大鼠血清和BALF中IL-8、TNF-α的含量,并通过H E染色和光镜观察气管、支气管及肺组织病理形态学改变。结果模型组大鼠支气管内有明显慢性支气管炎特征性病理学改变,肺组织显示肺泡结构紊乱,肺泡壁变薄或破裂,肺泡呈囊状扩张,部分融合成肺大疱,符合COPD形态学诊断指标,大鼠血清及BALF中IL-8、TNF-α的含量明显升高;与模型组比较,利金方高、低剂量组和补肺汤组COPD大鼠气管、支气管及肺组织病理学改变均有所减轻,血清及BALF中IL-8、TNF-α的含量均明显下降;利金方高剂量组的病理学减轻程度优于利金方低剂量组和补肺汤组;利金方高剂量组血清及BALF中IL-8、TNF-α的含量较补肺汤组均明显下降。结论利金方可以降低血及BALF中IL-8、TNF-α的表达,抑制气道炎症,从而达到治疗COPD的目的。 Objective: To observe the effects of liking prescription on IL - 8 and tumor necrosis factor - α(TNF - α) expression of chronic obstructive pulmonary disease (COPD) Qi Deficiency serum and bronchoalveolar lavage fluid (BALF). Methods: 50 SPF level Wistar rats were randomly divided into 5 groups: blank group, model group, the liking prescription high- dose group, the liking prescription low- dose group and Bufei Decoction groups. The rat models of COPD were established by complex factors. The drugs were administered to the appropriate dose for 14d. The IL - 8, TNF - α levels of serum and BALF were detected, and the morphological changes pathology were observed by HE staining and light microscopy. Results: The bronchia of model group rats showed obviously chronic bronchitis characteristic pathological changes in lung tissue that the structure of pulmonary disorder, alveolar wall thinning or rupture, was cystic dilatation of pulmonary, as part of the integration of lung bullae, in line with the morphology of COPD diagnosis. The IL - 8, TNF - α expression in BALF and serum increased significantly. Compared with the model group, the pathological changes of both liking prescription groups and Bufei Decoction group had reduced and IL - 8, TNF - α levels of serum and BALF were significantly decreased. The pathological changes and IL- 8, TNF-α levels decrease of liking prescription high- dose group were better than liking prescription low -dose group and Bufei Decoction group. Conclusion: Liking prescription can reduce the IL- 8, TNF- α expression in blood and BALF, and inhibit the airway inflammation, so as to achieve the treatment of COPD.
