摘要
目的筛选肝癌组织特异性癌症相关候选基因。方法应用基因芯片技术检测肝癌细胞系Bel7402-2、肾癌细胞系KETR、胃癌细胞系SGC基因及人胎肝细胞系L-02的全基因组序列,分析其基因表达差异。结果与肾癌细胞系KETR、胃癌细胞系SGC及人胎肝细胞系L-02相比,在检测的54614个探针组中,肝癌细胞系BEL7402-2表达共同上调2倍及2倍以上的有324个基因,表达下调2倍及2倍以上的有53个基因。涉及凋亡、细胞黏附、细胞增殖、信号通路等多种生物学过程。结论筛选出377个肝癌组织特异性及癌症特异性相关抗原,为进一步鉴定肝癌特异性诊断指标提供了侯选基因。
Objective To identify tissue-specific candidate genes associated with hepatocellular carcinoma(HCC). Methods The profiles of gene expression of hepatoma cell line Be17402-2 and renal carcinoma cell line, gastric carcinoma cell line, human fetal liver cells were analyzed by gene chips. Results Compared with renal carcinoma cell line, gastric carcinoma cell line, human the fetal liver cells,of the detected 54 614 probe sets, 324 genes were up-regulated by more than 2 folds and 53 genes were down-regulated by more than 2 folds in the hepatoma cell line Be17402-2, including the genes associated with apoptosis, cell adhesion,cell proliferation, and signaling pathway etc. Conclusion A total of 377 hepatoma tissue-specific and specific cancer-related genes have been screened, which provide candidate genes for further identifying specific diagnostic tumor markers for diagnosing HCC.
