摘要
目的观察棕榈酸棕榈酯对人外周血单核细胞增殖的影响。方法通过人淋巴细胞分离液及磁性细胞分选技术(MACS)分离纯化得到人外周血单核细胞,用不同浓度棕榈酸棕榈酯(0、1、2.5、5、20μM)干预,CCK8法检测人外周血单核细胞生长增殖的情况。结果棕榈酸棕榈酯能促进单核细胞的增殖。促增殖效应在20μM24h末达峰值,且棕榈酸棕榈酯浓度在0~20μM之间呈剂量依赖关系,在8~24h呈时间依赖关系(P<0.05)。结论棕榈酸棕榈酯能显著促进人外周血单核细胞的增殖,对人体可能具有毒性作用。
Obiective To explore the effect of palmityl palmitate on the proliferation of human peripheral blood monocytes. Methods The human peripheral blood monocytes were isolated by human lymphocytes separateness Ficoll-paque and magnetic activated cell sorting technique(MACS), which were stimulated with different concentrations of palmityl palmitate(0, 1,2.5,5,20 μM) and proliferation was detected with CCK8. Results Palmityl palmitate significantly promoted the proliferation of monocytes in a dose-and time-dependent manner, whose maximum effect appeared at 20 μM and 24 h (P〈0. 05). Conclusion Palmityl palmitate can induce the proliferation of monocytes, indicating that it may have toxicity for human.
