摘要
目的探讨钒联合维生素D3(VitD3)对结肠癌的治疗作用。方法100只裸鼠皮下接种HCT116结肠癌细胞,成瘤后原位移植,建立结肠癌裸鼠模型后随机等分为钒(A)组、VitD3(B)组、钒联合VitD3(C)组和对照(D)组。用药4周后,每组取18只脱颈处死,检测肿瘤体积和转移灶数量,免疫组化检测瘤体增殖细胞核抗原(PCNA)指数,其余裸鼠用于生存试验,流式细胞仪检测肿瘤细胞凋亡和细胞周期。结果C组裸鼠增殖指数显著低于D组(P<0.05),荷瘤裸鼠生存时间显著延长(P<0.05),肿瘤细胞凋亡增加(P<0.05),细胞停留在G0/G1期。结论钒联合VitD3具有明显的抗结肠癌细胞增殖、诱导细胞凋亡的作用。
Objective To study the effect of vanadium(V) combined with Vit D3 for the treatment of xenografted lolon cancer in nude mouse. Methods After establishment of the xenografted lolon cancer model, the mice were randomly divided into 4 groups with 25 mice each. The mice in group A were administered V(200 mg · kg^-1 · d^-1 )by lavage,in group B Vit D3 (2 U · kg^-1 · d^-1) by intramuscular injection, in group C V(200 mg · kg^-1 · d^-1) by lavage combined with Vit D3 (2 U · kg^-1 · d^-1) by intramuscular injection,and in group D normal sodium(200 ml · kg^-1 · d^-1) by intramuscular injection as the negative controls. Four weeks later, 18 mice from each group were killed,the gross tumor volume and numbers of metastatic nodules were measured. PCNA index of tumors were assayed by immunohistochemistry. The fast of mice in each group were used to evaluate the survival time. Apoptosis of tumor cells and tumor cells cycle were analyzed by flow cytometry when all mice died. Results Compeared with group D, the PCNA.index of tumor cells was less and survival time of mice was longer and apoptosis rate of tumor cells was increased in group C (P〈 0. 05). Tumor cells were arrested in G0/G2 phase as well. Conclusion Vanadium combined with Vit D3 significantly reduces the proliferation and induces the apoptosis of colon cancer cells.
