猪软骨多糖对S180荷瘤小鼠免疫功能的影响 被引量:3

Effect of the pig cartilage polysaccharide on the immunological function of S180 tumor-bearing mice

摘要 皮下注射S180细胞建立动物实体瘤模型,将小鼠随机分为生理盐水对照组和猪软骨多糖治疗组,连续腹腔注射生理盐水或猪软骨多糖21d,MTT法测定脾细胞增殖能力、免疫荧光法检测CD40和CD40L的表达以及TUNEL法检测瘤细胞的凋亡;免疫组化SP法检测Fas、PCNA、CyclinD1和p21的表达。结果显示,猪软骨多糖能够促进脾淋巴细胞的转化并提高CD40和CD40L蛋白的表达,治疗组的Fas和p21的抗体表达较对照组显著升高,而PCNA和CyclinD1的表达则显著降低。提示猪软骨多糖能够活化免疫细胞,提高小鼠的免疫力,通过提高Fas和p21的表达水平和降低PCNA和Cy-clinD1的表达水平而诱导瘤细胞的凋亡。 The solid tumor model of mice was established by subcutaneous injection of the S180 cells and this animal model was used for study on the effect of pig cartilage polysaccharide upon the immunological function of mice Mice bearing the S180 tumor were divided randomly into two groups, i.e. the treatment group with pig cartilage polysaceharide and saline control group. In both groups, mice were injected with pig cartilage polysaccharide or saline for successive 21 days. MMT colorimetry method was used to assay the multiplicative ability of spleen cells and the changes of CD40 and CD40L expressions were de tected by immunofluorescent assay. Meanwhile, the apoptotic rate of the tumor cells was determined by TUNEL technique and the expressions of Fas, PCNA, cyclin D1 and p21 were assayed by immunohistochemical SP method. It was demonstrated that pig cartilage polysaceharide enhanced the multiplicative ability of spleen cells and the expressions of CD40 and CD40L in comparison with those of the control mice. Compared with the control mice, the expressions of Fas and p21 was higher while those of PCNA and cyclin D1 was lower. These results indicate that pig cartilage polysaccharide can activate immune cells and enhance immunological function of mice, thus inducing apoptosis by increasing the expression of Fas and p21 and decreasing the expression of PCNA and cyclin D1 protein.
