摘要
利用生物信息学方法分析了西方蜜蜂Apis mellifera毒蕈碱型乙酰胆碱受体的核酸和氨基酸序列,并对其组成成分、疏水/亲水区、跨膜拓扑结构域、分子系统进化关系进行了预测和推断。结果显示,该受体定位在第8条染色体上,由618个氨基酸组成,分子量69906.5D,等电点(pI)8.56;是G蛋白偶联型受体,含N-糖基化位点、蛋白激酶C磷酸化位点、cAMP/cGMP依赖蛋白激酶磷酸化位点。
The Apis mellifera muscarinic acetylcholine receptor gene (XM 395760), which was registered in GenBank, was analyzed and predicted by the tools of bioinformatics in the following aspects: a composition of nucleic acid and amino acid sequence, equipotenfial, hydrophobidty or hydrophilidty, transmembrane topological structure, the comparison of homologous proteins and phylogenedc relationship. The results showed that Apis melhfera rnAchR gene was located at the chromosome 8, which had a putative protein of 618 amino acids with a predicted mol. wt. of 69 906.5 D and isoelectric point of 8.56, and that Apis mellifera mAchR belonged to a large family of membrane-bound receptors which were coupled to G-protein and included N- glycosylation sites, protein kinase C phosphorylation sites, cAMP-and cGMP-dependent protein kinase phosphorylation sites by PROSITE motifs.
