南海常见7种石斑鱼的DNA条码构建与亲缘关系分析 被引量:2

摘要 利用特异性聚合酶链式反应(PCR)技术扩增获取了南海海域常见的7种石斑鱼———鲑点石斑鱼(Epinephelus fario)、青石斑鱼(E.awoara)、黑边石斑鱼(E.fasciatus)、赤点石斑鱼(E.akaara)、七带石斑鱼(E.septemfasciatus)、斜带石斑鱼(E.coioides)、棕点石斑鱼(E.fuscoguttatus)———线粒体DNA COI基因及核DNA的RAG1基因部分序列。测定序列的比较分析结果显示:①该研究获取的COI基因部分序列能将7个物种分别开来,可以作为DNA条码;②线粒体DNA COI基因及核DNA的RAG1基因部分序列分别构建的系统发生树较为一致,结果都支持7种石斑鱼分为3支:青石斑鱼和斜带石斑鱼为第1支;鲑点石斑鱼、黑边石斑鱼、赤点石斑鱼和棕点石斑鱼为第2支;七带石斑鱼为第3支。研究表明,COI基因部分序列不但可以作为物种辨识的良好DNA条码,而且在石斑鱼属内的种间系统发生关系分析方面也具有一定的适用性。 PCR technology was used to get mitochondrial DNA COI gene and partial sequence of RAG1 gene of nuclear DNA for common seven kinds of Epinephelus in the south China sea, such as Epinephelus fario, Epinephelus awoara, E. fasciatus, E. akaara, E. septemfaseiatus, E. coioides and E. fuscoguttatus. The results showed that seven species could be separated by the partial sequence of acquired COI gene, and these sequences could be as DNA barcode, phylogenic trees based on COI and RAG1 gene were coincidence, they all supported the results that seven kinds of Epinephelus could be divided into three groups, the first group was E. awoara and E. coioides, the second was Epinephelus fario, E. fasciatus, E. akaara and E. fuscoguttatus, and the last was E. septemfasciatus. This study indicated that the partial sequence of COI gene not only was good DNA barcode to identify different species, but also had certain applicability in the analysis of interspecific phylogenetic relationships of Epinephelus.
