VEGF在大鼠急性缺血性肾功能衰竭的表达

Variations of levels of vascular endothelial growth factors in rats with acute ischemic renal failure

摘要 目的探讨建立大鼠肾脏缺血/再灌注损伤(ischemia reperfusion injury,IRI)模型后不同时段血管内皮生长因子(Vascular endothelial growth factor,VEGF)在血清及尿液中的浓度的变化及在ARF中的发病机制。方法建立大鼠肾脏IRI模型,采用双抗体夹心酶联免疫吸附法(ELISA法)检测IRI术后不同时间段血、尿VEGF水平,并与肾功能做相关分析。结果手术组术后6h血清尿素氮(BUN)、肌酐(SCr)、血钾显著升高,至术后48h达高峰,从术后72h开始回落(P<0.05);手术组术后6h血清VEGF显著升高,术后12h达高峰,从术后24h开始下降至正常;手术组术后6h尿液VEGF显著升高(P<0.01),至术后24h降至低于假手术组,从术后48h开始回升。结论模型大鼠血、尿VEGF水平在5~12h显著升高,可作为大鼠肾脏IRI早期肾脏缺血缺氧的监测指标。 Objective To observe the changes of vascular endothelial growth factor (VEGF) after setting up ischemia reperfusion injury (IRI) rat model. Methods Rat IRI model was established and the levels of blood, urine at various time priods after operation were determned with ELISA, renal function was also observed. Results The levels of BUN, SCr, blood patassium were enhanced and reached a peak 48 hours after operation, then they reduced 72 hours after operation (P〈 0.05). The levels of VEGF arised quickly 6 hours after operation and reached a peak 12 hours after operation, then it returned to noral 24 hours after operation. While the level of urine VEGF was increased rapidly (P〈0.01), then it reduced 24 hours after operation, then it enhanced 48 hour after operation. Conclusion The results show that the levels of BUN, SCr, blood potassium would rise after operaton at varous extent and with a peak occurred 48 hours after operation, indicating that injury to renal function most occurred 48 after operation. The ehancement of VEGF in blood and urine can be taken as an indicator for monitoring early IRI to the renal function of the rats.
