摘要
目的:观察联合应用奈达铂和放疗治疗荷Lewis肺癌实体瘤C57BL小鼠的疗效,以明确奈达铂对Lewis瘤的放射增敏作用。方法:建立小鼠Lewis肺癌实体瘤模型后,分对照组、单纯放疗组、奈达铂组及奈达铂联合放疗组。对照组给予腹腔注射生理氯化钠溶液0.1mL,单纯放疗组荷瘤鼠左前肢肿瘤局部以15GY射线照射,奈达铂组分2个亚组,分别以5和10mg·kg^-1奈达铂腹腔注射,联合放疗组则以5mg·kg^-1奈达铂腹腔注射加荷瘤鼠左前肢肿瘤局部15GY射线照射。然后每隔2d测量肿瘤的长、短径,比较各组瘤体积、肿瘤生长延缓天数、荷瘤鼠存活数和肺转移率。结果:与对照组比较,5和10mg·kg^-1的单剂量奈达铂都对Lewis实体瘤生长有抑制作用(P〈0.001);联合放疗组局部肿瘤生长受到明显抑制,抑瘤率为76.88%,明显高于单纯放疗组(57.21%,P〈0.001)和奈达铂组(23.57%,P〈0.001);联合放疗组肿瘤生长延缓天数(瘤体积1.1~2.5cm^3)超过10d,高于单纯放疗组(6d,P〈0.001)和奈达铂组(4d,P〈0.001);联合放疗组60d内存活的鼠最多(3个),单纯放疗组为1个,其他两组鼠全部死亡。联合放疗组平均肺转移为0.8个/叶肺,单纯放疗组为2.2个/叶肺,奈达铂组为6.04个/叶肺,对照组为20.2个/叶肺(前三者与对照组比较P〈0.001,联合放疗组与单纯放疗组比较P〈0.01)。结论:奈达铂可显著地增强小鼠Lewis肺癌细胞放射治疗敏感性,并减少肺转移。
Objective: To observe the effect of nedaplatin on radiosensitivity in mice bearing Lewis lung cancer. Methods: Mice bearing Lewis lung cancer were treated with radiation, nedaplatin or their combination The parameters of tumor growth, inhibition of growth, the number of surviving mice and metastasis were observed after the treatments. Results: Single dose of 5 or 10 mg. kg^-1 of nedaplatin significantly inhibited the growth of Lewis tumor (P 〈 0. 001 ). Tumor growth inhibition in combination group (76.88 % ) was higher than that of radia- tion alone (57.21% , P 〈 0.05 ) or nedaplatin alone (23.57 % , P 〈 0. 001 ). Time of tumor growth delay ( volume from 1.1 to 2.5 cm^3 ) was beyond 10 days in combination group, 6 days and 4 days in radiation alone and nedaplatin alone, respectively( P 〈0. 001 ). Three mice survived beyond 60 days in combination group, 1 in radiation alone, and none in other two groups. The number of lung metastatic cancer in combination group was 0.8 per lung lobe, which was fewer than those in control (20.2 per lung lobe, P 〈 0. 001 ) , radiation alone (2.2 per lung lobe,P 〈 0. 001 ) , and nedaplatin alone (6.04 per lung lobe, P 〈 0. 001 ). Conclusion: Nedaplatin can increase radiosensitivity of Lewis lung cancer and result in a significant inhibition of tumor growth and lung metastasis.
