摘要
目的:建立竹柏叶中德花双黄酮含量测定方法。方法:采用高效液相色谱法,色谱条件为:AgilentC18色谱柱(150mm×4.6mm,5μm);乙腈-0.05%磷酸水液为流动相,梯度洗脱(时间0~30min,乙腈15%~100%);流速为1.0mL.min^-1,检测波长为340nm。结果:穗花双黄酮在0.013—0.130μg范围内线性关系良好(r=0.9999),平均加样回收率为96.34%,RSD为1.4%(n=6)。结论:本法操作简便,结果准确,重现性好,可用于竹柏叶中穗花双黄酮成分的含量测定,并为其开发应用提供基础。
Objective : To establish a method to determine the content of amentoflavone in Podocarpus nagi leves. Methods: An HPLC method was carried out with an Agilent Hypersil ODS column (150 mm ×4.6 mm, 5μm). The mobile phases were acetonitrile (A) and 0.05% phosphoric acid solution (B) , and the gradient elution was performed in 0 -30 min with acetonitriie fiom 15% to 100%. The flow rate was 1.0 mL.min^-1 and the detection wavelength set at 340 nm. Results : The linear range of amentoflavone was within 0.013 - 0.130 μg. The average recovery rate was 96.34% with RSD of 1. 4% (n = 6). Conclusion: The method was simple, rapid, accurate and suitable for the detection of amentoflavone in Podocarpus nagi leves. The results provided a basis for the development and application of Podocarpus nagi leves.
