Rotavirus double-stranded RNAs(dsRNA) isolated from kid stool samples were reverse transcripted and amplified by PCR using two oligonucleotide primers derived from the genome 3 fragment of IDIR strain of group B rotavirus (GBRV).The 290-bp amplified products were labeled with photobiotin as probe for the detection of GBRV in stool specimens by dot-blot hybridization.The results suggested that genomic RNA of GBRV strain of lamb,kid and calf were readily detected with strong positive signals by hybridization,whereas the samples from fecal specimens containing porcine and ovine GARV,porcine GBRV,GCRV or fecal specimens from uninfected controls gave negative reaction.The PCR method provided a rapid and efficient mean by which large quantities of cDNA suitable for probing to detect GBRV could be obtained.
