摘要
从野生型甘蓝黑腐黄单胞菌中筛选出XCCNAU—01作为育种的出发菌株。用硫酸二乙酯和氯化锂复合诱变得系列突变体,经平板、摇瓶发酵必传代试验筛选出黄原胶生产菌株XCCNAU-92。对蔗糖、玉米淀粉等碳源皆有较强的利用能力,当以蔗糖为碳源时,30℃,102r/m发酵72h,产胶量达36.25g/kg,提高12.9%,粘度达17200cp,增加43.3%,碳源转化率达72.5%。
The XCCNAU - 01 strain was selected from the wild type Xanthomonas camp stris PV. campestrts as parent strain of breeding, many mutants were obtained by treating the XCCNAU - 01 strain with dirtyl sulfate and LiCl, the xanthan gum producing strain XCCNAU - 92 selected by plate, shake flask culture and generation experiment could make full use of surcose, maize starch etc. Surcose utilized as carbon source, under·30℃ 120r/m. fermentative period 72 hours, the yield of the XCCNAU -92 is 36- 25g/kg, more than that of the parent strain by 12.9 percent, the visosity of fermented mash is 17200cp, raising by 43.3 percent. the transformation ratio of carbon source is 72.5 percent.
