摘要
利用ISSR分子标记对36种冬青属植物的种间关系进行了研究,从100条引物中筛选出12条能扩增出清晰带型并具多态性的引物,共扩增出235条DNA片段,相对分子质量主要集中在400—1500bp之间,其中232条片段呈多态性,多态率为98.7%,平均每条引物扩增出19.6条DNA片段;依据扩增结果用UPMGA类平均聚类法进行遗传相似系数分析并构建了系统树.结果表明,36种冬青属植物在遗传相似系数O.64处明显聚为落叶类和常绿类,初步证明了利用ISSR分子标记对冬青属植物进行系统进化和亲缘关系分析的可行性.
Inter. species relationship of 36 species in llex was studied by ISSR markers. 12 primers were screened out from 100 random primers. 235 DNA bands were amplified ranging from 400 - 1 500 bp, the polymorphic rate of 232 bands accounted for 98.7%. The average number of DNA bands amplified by each primer was 19.6. Genetic similarity coefficient was calculated by NTSYS and the dendrogram were constructed by UPMGA. 36 Ilex plants were classified into deciduous plant species and evergreen plant species at similarity coefficient 0.64. The preliminary proof proved that it was feasible to analyse the system evolution and genetic relationship of flex plants species by using ISSR.
