摘要 目的:建立比格犬血浆中丹参酮ⅡA的测定方法。方法:以地西泮为内标,采用HPLC-MS/MS方法,以Luna Phenyl(30mm×2.0mm,3μm)色谱柱为分析柱,5mmol·L-1甲酸铵缓冲液(用色谱纯甲酸调pH至3.0±0.05)-甲醇(含0.1%的甲酸)为流动相,梯度洗脱,流速为0.2mL·min-1,柱温为25℃,进样量为20μL。结果:在1~1000ng·mL-1浓度范围内具有良好的线性关系,线性方程为:Y=0.00519X-0.00439,r=0.9980(n=8)。准确度、精密度均符合生物样品的测定要求,最低定量浓度为1ng·mL-1。结论:该检测方法简便、准确,专属性强,能够满足丹参酮ⅡA在比格犬体内药代动力学研究的需要。 Objective :To Diazepam was used as the develop a method for determination of tanshinone IIA in plasma of Beagle dog. Method: internal standard. Tanshinone IIA was chromatographed on Thermo - Finnigan LCQ Quanturn with a Luna Phenyl column (30 mmx 2.0 mm, 3 μm). The mobile phase was 5 mmol·L^-1 ammonium formate solution(using formic acid with HPLC grade to adjust to pH 3.0±0.05) -methanol with 0. 1% formic acid at the flow rate of 0. 2 mL·min^- 1 ( gradient elution) , the column temperature was 25℃ and the injection volume was 20μL. Result: A good linear relationship was obtained between the peak area ratio of tanshinone IIn/diazepam and the concentration of tanshinone IIA over the range of 1 to 1000 ng·mL^-1. The linear equation was Y = 0. 00519X - 0. 00439, r = 0. 9980 ( n = 8 ). The limit of quantitation was 1 ng·mL^-1. Conclusion: The method is simple, fast, accurate and suitable for the study of pharmacokineties in Beagle dog.
