HPLC determination of nuezhenide in Fructus Ligustri Lucidi

目的:建立女贞子药材的含量测定方法。方法:采用高效液相色谱法测定女贞苷的含量。使用Alltima C18色谱柱(4.6mm×250mm,5μm),流动相为乙腈(A)-水(B),梯度洗脱[0～40min,15%A→40%A],流速1.0mL·min-1,检测波长230nm。结果:女贞苷线性范围50～1000μg·mL-1(r=0.9999);平均回收率(n=5)为99.01%(RSD=0.17%)。不同来源10批女贞子药材含量测定结果表明,女贞子药材中女贞苷含量为0.24%～3.06%,综合分析10批样品含量测定结果,建议女贞子药材中含女贞苷应不低于1.7%。结论:本方法简便、准确,重复性好,专属性强,可作为女贞子药材质量控制方法。

Objective：To develop a quantitative method for the determination of nuezhenide in Fructus Ligustri Lucidi. Method：An HPLC method was applied to separate and quantify nuezhenide. The samples were separated by an Alhima C18column（4.6 mm×250mm,5μm） with acctonitrile（A） -water（B） as mobile phase[0-40 rain,15% A→40% A]. The flow rate was 1.0 mL·min^-1 ,and the detection wavelength was set at 230 nm. ResuLts：The linear range of nuezhenide was 50 - 1000 μg·mL^-1 （ r = 0. 9999）, and the average recovery （ n = 5 ） was 99.01% （RSD = 0.17% ）. As the results shown,the content of nuezhenide in Fructus Ligustri Lucidi was 0. 24% -3.06% from 10 different habitats. Our study suggests that the content of nuezhenide in Fructus Ligustri Lucidi should not be less than 1.7%. ConcLusion ： The developed method is sensitive, rapid, accurate and specific. And it can be used to control the quality of Fructus Ligustri Lucidi.
