豆渣中可溶性大豆蛋白肽的提取及结构表征 被引量:7

摘要 研究了豆渣的预处理条件,采用正交实验设计法确定了蛋白酶水解豆渣制备可溶性蛋白肽的最佳工艺,并对其结构进行表征。结果表明:豆渣在120℃、pH9·0条件下预处理15min,可明显增加可溶性蛋白肽的提取率。先采用碱性蛋白酶在pH9·0、温度55℃条件下作用4h后,再用木瓜蛋白酶在pH7·0下作用4h,可溶性蛋白肽提取率达到了63·03%,氨基酸态氮含量控制在5·15%;傅里叶变换红外光谱分析表明,该可溶性大豆蛋白肽具有蛋白质和多肽的双重特征。 The extraction of soluble soybean protein peptides from soybean dregs with enzymatic method was studied.The condition of pretreatment of soybean dregs was determined.Based on the result, the orthogonal experiment was designed to obtain the optimum process conditions of preparing soluble soybean protein peptides, and the hydrolyzation technology of extraction of soluble soybean protein peptides by protease proteinases and papain was established .The result showed that it was best for the extraction of soluble protein peptides from soybean dregs, which was dealed with in the condition of temperature 120℃ and pH9.0 for 15min.The soybean dregs were hydrolyzed by protease proteinases in the condition of pH9.0,55℃ for 4 hours.After this,they were hydrolyzed continuously by papain in the condition of pH7.0,55℃ for 4h.Under this condition,the rate of soluble protein peptides extraction was 63.03% ,the amount of ammonia nitrogen was 5.15%.FT-IR analysis showed that soluble soybean protein peptides meet dual characteristics of protein and peptides.
