银杏叶对大鼠肾脏中组织型转谷氨酰胺酶的表达及干预作用 被引量:2

摘要 目的:探讨组织型转谷氨酰胺酶(tTG)在单侧输尿管梗阻(UUO)大鼠肾间质中的表达及银杏叶(GBE)对其表达的影响。方法:采用左侧输尿管结扎术复制肾间质纤维化模型,将雄性W istar大鼠随机分为假手术组、模型组、贝那普利(15 mg.kg-1)治疗组及银杏叶(300 mg.kg-1)治疗组。术后第14天处死各组大鼠,取左肾进行HE和M asson染色,观察肾间质纤维化病变;免疫组化法检测肾间质中纤维连接蛋白(FN)和tTG的表达;RT-PCR检测tTG mRNA表达。结果:模型组FN和tTG的表达明显高于假手术组(P<0.01);贝那普利组和银杏叶组治疗后二者显著低于模型组(P<0.05)。结论:贝那普利及银杏叶制剂可能通过下调tTG而减轻肾间质纤维化。 Objective: To investigate the expression of tissue transglutaminase (tTG) in unilateral ureteral obstruction (UUO) rats and the intervention of Folium Ginkgo(GBE). Method: The animal models of unilateral ureteral obstruction (UUO) were used. Wistar rats were randomly divided into four groups, the Sham-operated group, the UUO group, the benazepril (Bena)-treated UUO group and the GBE-treated group. The rats were sacrificed at day 14. Histological changes in renal tubular interstitium were observed with HE and Masson staining, and the mRNA and protein levels of tTG and FN were detected by RT-PCR and immunohistochemistry. Result: Compared with the sham group, the expression of tTG and FN were significantly increased (P 〈 0. 01 ), and decreased after been treated by Bena and GBE ( P 〈 0.05 ). Conclusion : Bena and GBE may suppress the development of fibrosis partially via downregulation of tTG expression.
