一种简单高效提取棉花不同组织总RNA的方法

A Simply and High Efficiency Method of Total RNA Isolation in Upland Cotton

目前,虽然已经报道了几种比较有效的提取棉花组织总RNA的方法,但是这些方法往往费时费工,又很难同时适用于棉花不同组织的总RNA提取。以CTAB法为基础进行技术改良,同时结合LiCl沉淀等,总结出了一种简单、快速又普遍适用于棉花各组织总RNA的提取方法。试验证明,此法提取到的RNA纯度高、完整性好,适用于逆转录合成cDNA第一链、RT-PCR分析以及Northern杂交等分子实验。

At present several methods of cotton RNA isolation were reported, but they were time and labor consuming, and incompatible for all cotton tissues. Based on the CTAB extraction method and the LiCI precipitation method for total RNA, a fast and simply method for cotton total RNA isolation was developed, which is also suitable for most of cotton tissues. The cotton total RNA produced by this method was high purity, integrality, and could be used in reverse transcription expression analysis, RT-PCR analysis and Northern blotting etc.
