摘要
[目的]研究X线照射对鼻咽癌细胞株CNE1、CNE2中ATM mRNA表达水平的影响。[方法]采用逆转录酶聚合酶链式反应(RT-PCR)技术,以β-actin为内参照,测定10GyX线照射前后CNE1、CNE2中ATM mRNA表达水平的变化。[结果]在X线照射前,CNE1中ATM mRNA基础水平与CNE2相似;在X线照射后,CNE1中ATM mRNA水平在照射4h后明显升高,12h后恢复;CNE2中ATM mRNA水平在照射后12h内无显著变化。[结论]在具有不同放射敏感性的鼻咽癌细胞株CNE1、CNE2中,ATM mRNA表达的基础水平相似,在经X射线处理后,CNE1和CNE2中ATM mRNA水平变化规律不同。这种ATM mRNA表达差异可能是导致两者放射敏感性不同的原因之一。
[Purpose] To study the effect of X-irradiation on the expression of ATM mRNA nasopharyngeal carcinoma (NPC) cell line CNE1 and CNE2 with different radiosensitivity. [Methods ]Reverse transcriptase polymerase chain reaction (RT-PCR) was used to measure the changes of ATM mRNA levels before amt after 10Gv X-ray irradiation in NPC cell lines CNE1 and CNE2 with β-actin as internal reference. [Results] Before irradiation, the basic ATM mRNA levels in CNE1 was similar with those in CNE2: after irradiation, the ATM mRNA levels was significantly increased at 4th hour in CNE1 and recover' at the 12th hour. The ATM mRNA levels was no significant change in CNE2 within 12 hours irradiation. [Conclusion] The basic ATM mRNA levels are similar in NPC cell lines CNE1 and CNE2 with different radiosensitivity. After X-ray irradiation. the variation of ATM mRNA levels was different in CNE1 and CNE2 might be one of the reasons of different radiosensitivity.
