摘要
草鱼CCL4基因全长854 bp,最大开放阅读框位于104~397 bp,编码97个氨基酸,其中32~73氨基酸为其保守的CC趋化因子家族结构域,N-端有1个由24个氨基酸组成的信号肽。草鱼CCL4与斑马鱼的同源性为69%,与其他鱼类和哺乳类的同源性在50%以下。系统进化分析也显示其与斑马鱼CCL4基因的亲缘关系最近。CCL4基因在草鱼各组织中都有表达,以脾脏表达量最高,心、肌、鳃、肝中次之,肠和肾中有少量表达,在脑中有痕量表达。经Poly I:C感染后,在肝脏、脾、肠、肾、鳃中,CCL4的表达水平有不同程度的增加,而在心、肌、脑中无明显变化。
The full - length of CCL4 in grass carp is 854 bp. The largest ORF encods 97 amino acids protein, with the conservative CC chemokine family domain located at 32 -73aa and a signal peptide of 24 amino acids in N - terminal. Grass carp CCL4 shared the highest homology of 69% with zebrafish CCL4, and the homology with catfish, atlantic salmon,rainbow trout and so on was less than 50%. Phylogenetic analysis also showed the closest relation- ship with zebrafish CCL4. CCL4 is expressed in all detected tissues of grass carp, there is the highest expressional level in spleen, followed by heart, muscle, gill, liver, little amount in intestine and kidney, and trace amount in brain. The expression in liver, spleen, intestine, kidney and gill increased at different degrees after infected by Poly I:C ,while no significant change was observed in heart, muscle and brain.
