脑缺血再灌流后大鼠海马锥体细胞和星形胶质细胞的体视学 被引量:1

The Stereological Study on Pyramidal Cells and Astrocytes in Rat Hippocampus after Cerebral Ischemia-Reperfusion

摘要 为了研究脑缺血再灌流后海马锥体细胞和星形胶质细胞的变化,选取24只SD大鼠,随机分为对照组、缺血再灌流后6、12、24 h组,采用Nissl染色,GFAP免疫组织化学方法并应用体视学参数分别对海马CA1、CA3区锥体细胞和星形胶质细胞进行形态学定量分析。结果显示,海马CA1、CA3区锥体细胞体积密度(Vv)和表面积密度(Sv)随着缺血再灌流后存活时间的延长而减小,与对照组相比,6 h组显著降低(P<0.05),12 h组和24 h组极显著降低(P<0.01);而海马区星型胶质细胞的Vv、Sv却随缺血再灌流后存活时间的延长而加大,与对照组相比,6 h组无显著性差异(P>0.05),12、24 h组具极显著差异(P<0.01,P<0.01);同组海马CA1和CA3区,以及同组同一海马区域左右两侧比较,锥体细胞和星形胶质细胞Vv、Sv均无显著性差异(P>0.05)。以上结果表明,海马CA1、CA3区锥体细胞随着缺血再灌流时间的延长受损加重,而星形胶质细胞功能趋于活跃且与锥体细胞损伤密切相关。 To investigate the changes of pyramidal cells and astrocytes in rat hippocampus after ischemia - reperfu- sion ,24 SD rats were randomly divided into four groups which were control group, ischemia -reperfusion 6 h group, 12 h group and 24 h group. Changes in hippocampus CA1, CA3 were observed by Nissl staining and anti - GFAP im- munohistochemistry, stereological method was used to evaluate changes as well. The results showed that the volumn density (Vv) and surface density (Sv) of pyramidal cell in CAl and CA3 decreased as reperfusion time went on, and reperfusion 6 h, 12 h,24 h groups had distinct differences compared with control group (p 〈0.05 ,p 〈0.01 ). While the Vv and Sv of astrocytes increased with the signifi cant difference ( P 〉0.05 ), 12 h ,24 h group distinct differences between CA1 region ( p 〉 0.05 ). These results reperfusion time, compared with control group ,6h group had no had significant difference (p 〈0.01 ,p 〈0.01 ). There weie no and CA3 of the same group as well as between the left and right side of the same indicated that neuron damage became more severe as reperfusion time extended, while astrocytes functioned more actively and were closely related with neurons damage degree.
