摘要
采用离体心脏灌流及冠脉结扎方法,复制大鼠心肌缺血再灌注损伤的模型,观察丹参素对缺血及再灌性心律失常的保护作用。实验表明,离体灌注大鼠缺血期心律失常的发生率为85.7%,主要为传导阻滞及室性早搏和室性心动过速,但无室性纤颤发生;再灌期心律失常发生率高达100%,主要为室性早搏和室性心动过速,室性纤颤发生率为57.1%。0.5μmol/L异搏定和4mg/L丹参素均能使大鼠缺血再灌性心律失常发生率明显降低、持续时间缩短,但以丹参素的预防效果更佳。上述剂量的异搏定和丹参素,分别使缺血性心律失常发生率降低了33.3%和50.0%;再灌性心律失常发生率降低了71.4%和85.7%(P分别<0.05和0.001)。
This paper investigated the protecting effects of DS 182 on the ischemia/reperfusion arrhythmias in the isolated rat using the Langendorff method and ligation of coronary artery.The results of the experiment showed that the incidence of arrhythmias in the ischemic period and the reperfusion period was 85.7% and 100.0%,respectively.Administration of verapamil (0.5μmol/L) and DS 182(4 mg/L)to the perfusate significantly reduced the incidence and the onset of ventricular arrhythmias,but the preventing action of DS 182 was more efficient than verapamil.Verapamil and DS 182 reduced the incidence of ischemic arrhythmias by 33.3%and 50.0%,the incidence of reperfusion arrhythmias by 71.4%and 85.7%,respectively (P<0.05 and 0.001,respectively).It was demonstrated that Danshen products have significant preventing effect on ischemia/reperfusion arrhythmias.
