老年鼠脑老化神经细胞凋亡及基因调控的研究

Study on apoptosis of neural cell and its gene regulation in brain of aging rats

目的研究老年鼠脑老化神经细胞的凋亡及基因调控。方法用ＴＤＴ法及流式细胞仪观察不同鼠龄海马神经细胞凋亡及凋亡发生率，并用免疫组织化学检测ｂｃｌ－２、ｂｃｌ－ｘ１及ｂａｘ基因表达。结果老年组神经细胞在荧光显微镜下显示较典型的细胞凋亡特点，老年组海马神经细胞凋亡率与青年组比较有显著差异。老年鼠ｂｃｌ－２，ｂｃｌ－ｘ１表达下调，而ｂａｘ的表达量明显增加。结论脑老化过程中存在着以细胞凋亡形式的神经细胞的死亡。

おim:To study the apoptosis of neural cells and its gene regulation in brain of aging rags.Methods:To identify the morphological characters and percent of neuronal apoptosis in hippocampus with in TDT(teminal deoxynuleotidyl transferasemediated nickend labeling) and FACS can to determine the expression of bcl2,bclx1,and bax by immunohistochemical staining.Results:There is typical characters of neuronal apoptosis in old group.The percent of neuronal apoptosis of old group is higher than that yourger group(P<001).The expression of bcl2,bclxl decreased and bax incressed in old group.Conclusion:there is neuronal apoptosis in aging of the brain and the change of neuronal apoptosis consisitant with gene expression.
