摘要
目的探讨血浆纤维蛋白原(Fbg)Bβ-455G/A、-854G/A基因型的分布特征及其基因多态性联合多种生理和环境因素对Fbg浓度和分子活性的影响。方法采用整体抽样的方法选取开滦集团职工1555人,样本均清晨空腹抽取静脉血测定血糖等12项生化指标;应用聚合酶链反应-限制性酶切法进行两位点的基因多态性分析;采用微机辅助血浆Fbg功能自动监测系统测定血浆Fbg浓度和Fbg单体聚合反应速率(FMPV)、最大光密度(Amax)、FMPV/Amax等反映Fbg分子聚合功能参数;并进行体检和问卷调查。结果Bβ-455等位基因G、A频率分布为0.798、0.202,Bβ-854等位基因G、A频率为0.899、0.101。两位点基因连锁不平衡分析为随机分布,两位点等位基因及基因型频率分布与性别无关(P>0.05);而高血压病组Bβ-455等位基因G、野生基因型频率均高于无高血压病组(P<0.05);Bβ-455各基因型组间Fbg浓度及分子活性指标均无差异性(P>0.05),Bβ-854GA+AA组Fbg浓度、FMPV/Amax、FMPV及Amax均明显高于GG组(P<0.01)。结论Bβ-455等位基因G和GG基因型与高血压病有关,但对Fbg浓度和分子活性无明显影响;Bβ-854的变异基因型可以明显影响血浆Fbg浓度和分子聚集活性,但可以通过改变某些生理和环境因素增强或减弱其影响的程度。
Objective To investigate the genotype distribution of β-Fbg gene-455G/A,-854G/A, and gene Bβ-455G/A,-854G/A polymorphisms combined many physiological and environmental factors electing on Fbg concentration and molecular reactivity. Methods 1555 subjects from Tangshan coalminewere enrolled with study method of cluster sampling. The Bβ-455G/A,-854G/A polymorphisms were detected by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP). Fbg concentration,fibin monome polymerized velocity ( FMPV), absorbance maximum ( Amax), FMPV/Amax and twelve biochemistry factors including glucose were measured. Face to face interviews were conducted to fill in questionnaires by trained interviewers. Results The frequencies of the G-455, A-455, G-854, A-854 allelewere 0. 798,0. 202, 0. 899,0. 101, respectively. The relationship between Bβ-455G/A and Bβ-854G/A was a random distribution. No different prevalence rates of Bβ-455G/A and -854G/A were found on gender(P 〉0.05) ,but the distribution of G455 allele and GG genotype in the hypertension were higher than without the hypertension ( P 〈 0.05 ). No difference of Fbg concentration and molecular reactivity in Bβ- 455 G/A genotypes was found ( P 〉 0.05 ), The level of Fbg, FMPV/Amax, FMPV and Amax in GA and AA combined genotyp was significantly higher than that in GG genotype of Bβ-854 gene (P 〈 0.01 ). Conclusion G-455 allele andGG genotype of Bβ-455 have related in hypertension, but there is no association between Bβ-455 and Fbg concentration, molecular reactivity; Bβ- 854 genotypes have impact on Fbg concentration and molecular reactivity, further more, physiological and environmental factors might have coordinating functions.
