摘要
目的分析丹参酮ⅡA对免疫性血管炎、血小板及凝血活性的影响,探讨其抗血小板、抗凝及降低免疫性血管炎炎症反应的机理。方法建立免疫性血管炎模型,检测外周血小板数量、血小板聚集功能,流式细胞仪检测血小板活性标志Annexin V,ELISA法检测凝血酶原片断F1+2,HE染色、弹力纤维染色及电镜观察血管病理改变。并设立正常、模型和阿司匹林对照组。结果免疫性血管炎急性期血小板数量异常增高,血小板聚集功能、Annexin V检测的血小板活性以及凝血酶原片断F1+2活性也有明显增高。丹参酮ⅡA治疗后,血小板数量明显降低,为931.33±254.19×109/Lvs544.00±83.88×109/L,差异有统计学意义。Annexin V检测的血小板活性以及凝血酶原片断F1+2活性也明显降低,分别为22.55±6.00%vs12.27±6.25%、215.93±66.04pmol/Lvs148.69±14.40pmol/L,差异有统计学意义。与阿司匹林作用相比,除抑制血小板聚集作用稍弱外,其他作用两种药物差异均无统计学意义。结论丹参酮ⅡA可能通过降低血小板活性及凝血活性从而降低免疫性血管炎的炎症反应,减轻血管的病理损害,达到治疗的目的。
Objective To study the effects of tanshinone Ⅱ A on platelets and coagulation in immune vasculitis. Methods The model of immune vasculitis of rabbits were reproduced by intravenous injection of bovine serum albumin twice. Periphery platelet number, platelet aggregation and the population of Annexin V in platelets were detected. Prothrombin fragment F1 + 2 was dectected by ILISA method. The Pathological changes of immune vasculitis were analyzed by hematoxylin and eosin staining, elastic fibers staining and electron microscopy. Results The platelet number, platelet aggregation, the population of Annexin V and the lev- el of prothrombin fragment F1 + 2 at the acute phase significantly increased in immune vasculitis model group than those in control group. Tanshinone Ⅱ A significantly decreased platelet number from 931.33 ±254.19 × 10^9/L to 544.00 ± 83.88 × 10^9/L. The population of Annexin V and the level of prothrombin fragment F1 + 2 also significantly desreased from 22.55 ±6.00% to 12.27 ±6.25% ,215.93 ±66.04 pmoL/L to 148.69 ± 14.40 pmol/L. The inhibition effect of tanshinone Ⅱ A was as the same as aspirin except for the inhibition to platelet aggregation. And both drugs could improve the vessels damage of immune vasculitis. Conclusions Tanshinone Ⅱ A might improve the inflammation damage of vessels in immune vasculitis through the inhibition of platelets and coagulation activity.
