PPAR-γ_2在体质量正常的非酒精性脂肪肝患者腹部皮下和网膜组织中的定量表达

摘要 目的探讨过氧化物酶体增殖物激活受体γ2(PPAR-γ2)基因在体质量正常的非酒精性脂肪肝(NAFLD)患者皮下(SC)和网膜(OM)脂肪组织中的表达及其与胰岛素抵抗指数(HOMA-IR)的关系。方法应用SYBRGreen I实时定量逆转录聚合酶链反应(RT-PCR)法检测26例体质量正常的非酒精性单纯性脂肪肝患者(NAFLD)和26例体质量正常非NAFLD患者对照组腹部皮下(SC)和网膜(OM)脂肪组织PPAR-γ2mRNA的表达水平。结果(1)NAFLD组OM脂肪组织的PPAR-γ2mRNA相对表达量低于SC脂肪组织(t=-1.0,P=0.000 2);对照组两者间表达差异无统计学意义(t=0.30,P=0.77)。(2)NAFLD患者OM脂肪组织PPAR-γ2mRNA表达量低于对照组(t=-3.36,P=0.002),而SC脂肪组织PPAR-γ2mRNA表达量则高于对照组,但差异无统计学意义(t=1.80,P=0.08)。(3)相关分析表明NAFLD组HOMA-IR与OM脂肪组织PPAR-γ2mRNA表达呈负相关(r=-0.75,P<0.01),与SC脂肪组织PPAR-γ2mRNA表达(r=-0.57,P<0.01)呈正相关。结论NAFLD患者OM和SC脂肪组织PPAR-γ2mRNA表达异常,且与IR有关。 Objective To investigate the expression of peroxisome proliferators-activated receptor γ2 (PPAR-γ2) in abdominal subcutaneous (SC) and omental (OM) adipose tissue of normalweight nonalcoholic simple fatty liver disease (NASFLD) patients and its relation with insulin resistance (HOMA IR). Methods 26 normal-weight NASFLD patients were randomly selected as NASFLD group (n=26),and 26 normal-weight non-NASFLD patients were selected as control group (n=26). SC and OM adipose tissues were obtained from both groups. PPAR-72 mRNA expression levels in SC and OM adipose tissue were quantified using SYBR Green I real-time RT- PCR. Results (1) In NASFLD group,the PPAR-γz mRNA expression level in OM adipose tissue was significantly lower than that in SC (t=-1.0,P=0. 000 2);while in control group,there was no significant difference in PPAR-γz expression between OM and SC (t = 0.30, P = 0.77). (2) The PPAR-γ2 mRNA expression level in OM adipose tissue of NASFLD group was significantly lower than that of the control group (t=-3.36,P=0. 002). However,the PPAR-γ2 mRNA expression level in SC adipose tissue of NASFLD group was higher than that of control group without significant difference (t = 1.80, P = 0.08). (3) In NASFLD group, The HOMA-IR was negetively correlated with the PPAR-γ2 mRNA expression level in OM adipose tissues(r =-0. 75,P〈 0.01),and was positively correlated with that in SC(r = -0. 57, P〈0.01. Conclusion The expression level of PPAR-72 mRNA in OM and SC adipose tissue of NASFLD patients is abnormal and is related to IR.
