摘要
目的从细胞凋亡的角度研究骆驼蓬总碱(TAH)抗肿瘤机制。方法将TAH以不同剂量和作用时间处理体外培养的人宫颈癌HeLa细胞,用流式细胞仪、电子显微镜、DNA琼脂糖电泳检测TAH诱发HeLa细胞凋亡情况。结果一定剂量TAH作用一定时间后,HeLa细胞的流式细胞仪直方图出现亚二倍体峰(Ao峰);电子显微镜检测到染色质边集、浓缩,在核膜下形成小团块以及胞浆起泡,凋亡小体形成等一系列凋亡的特征性变化;DNA琼脂糖电泳图上出现DNA梯形电泳(DNAladder)。结论在一定剂量和时间范围内,TAH能诱导HeLa细胞凋亡,诱导细胞凋亡可作为TAH抗肿瘤的机制之一。
bjective The study was carried out to determine the mechanism of TAH (Total Alkaloid of Harmaline). Methods Treated with different TAH concentrations for different times,HeLa cells were tested by flow cytometry,DNA agarose electrophoresis,light microscopy and transmissional electron microscopy. Results In a certain range of TAH concentration and treatment time,the cells were characterized by the occurrence of subdiploid peak (Ao peak) on the histogram of flow cytometry and DNA ladder on the agarose electrophoresis.Transmissional electron microscopy showed that there were marginization of nuclear chromatin,formation of capshaped structures underlying the nuclear envelope,protruding of cell surface and formation of apoptotic bodies. Conclusion TAH could induce apoptosis in HeLa cells.
