罗格列酮对大鼠AMI再灌注后无再流的影响与VCAM-1、E-选择素的表达调控

摘要 目的观察罗格列酮对大鼠急性心肌梗死(AMI)再灌注后无再流的影响,并探讨该作用与血管细胞黏附分子-1(VCAM-1)、E-选择素表达调控的关系。方法健康成年SD大鼠36只,随机分为假手术组、对照组、罗格列酮组各12只;结扎冠状动脉左前降支3h继而松解2h建立无再流动物模型;再灌注后2h以病理染色法检测无再流范围;免疫组化、免疫荧光组化、RT-PCR等方法检测VCAM-1、E-选择素蛋白及mRNA的表达;Western blotting检测磷酸化κB抑制蛋白α(p-IκBα)。结果与对照组比较,罗格列酮组无再流范围显著减小(P<0.05),VCAM-1、E-选择素免疫组化染色强度及mRNA表达均明显减弱(P<0.05),p-IκBα水平显著降低(P<0.05)。结论罗格列酮显著减轻大鼠AMI再灌注后无再流范围。 Objective To investigate the effect of rosiglitazone on no-reflow after reperfusion in rats with AMI, and if the effect of rosiglitazone is related to the regulation of vascular cell adhesion molecule-1 ( VCAM-1 ) ,E-seleetin. Methods 36 healthy adult SD rats were randomly divided into the sham operated group, the control group, rosiglitazone group; To establish the animal model of no-reflow, left anterior descending coronary arteries were occluded for 3h and reperfusion for 2h;Pathological staining for determination of area of no-reflow;Immunohistochemical staining ,immunofluorescent staining ,RT-PCR for proteins and mRNA expression of VCAM-1 and E-selectin, Western blotting for level of phosphorylated inhibitory of KB p-IKBα were completed ; Results Compared with the control group, not only ANR% in rosiglitazone group was significantly reduced ( P 〈0.05 ) ;but also proteins and mRNA expression of VCAM-1 and E-selectin,level of p-IKBα protein were significantly de- creased (P 〈 0.05 ) ;Conclusion Rosiglitazone can significantly reduce the area of no-reflow after reperfasion in rats with AMI.
