摘要
目的:探讨表没食子儿茶素没食子酸酯(EGCG)对乳腺癌细胞MCF-7生长的影响及对乳腺癌细胞MDA-MB-231迁移的影响。方法:MCF-7细胞培养贴壁之后,加入EGCG处理,2d后收集蛋白,采用Western Blot检测磷酸化p38丝裂原活化蛋白激酶(phospho-p38MAPK)的表达;同样处理后收集活细胞,用细胞计数法检测细胞的存活;取对数生长期的MDA-MB-231细胞,分至6孔板培养,使用EGCG处理后,采用细胞划线法探测乳腺癌细胞的迁移。结果:使用EGCG处理乳腺癌细胞后,phospho-p38MAPK的表达降低,EGCG处理乳腺癌细胞4d后其增殖率降低50%,迁移活性降低。结论:EGCG处理乳腺癌细胞能抑制肿瘤细胞的生长以及迁移,这与p38 MAPK信号通路相关。
Objective: To determine the roles of epigallocatechin-3-gallate (EGCG) in the progression and migration of breast cancer cell lines MCF-7 and MDA-MB-231. Methods: MCF-7 cells were cultured and then treated with EGCG, Western Blot was used to detect the expression of phospho p38 mitogen-activated protein kinase (phospho-p38MAPK), and the cell survival was detected with cytometry. MDA-MB-231 cells were treated with EGCG, and its migration ability was detected by migration ability test. Results: When treated with EGCG, phospho-p38MAPK expression was abrogated, breast cancer cell's progression was down-regulated by 50%, and tumor cell's motility and migration potential was weakened. Conclusion: EGCG can inhibit breast cancer cell's progression and migration potential, and this is mediated by p38MAPK signaling pathway.
