RT-PCR技术检测石蜡包埋恶性生殖细胞肿瘤组织中Oct-4的表达

To examine the expression of Oct-4 from the paraffin wax Embedding Malignant germ cell tumors tissue with RT-PCR

目的:用逆转录PCR技术(RT-PCR)技术从石蜡包埋的恶性生殖细胞肿瘤组织中检测Oct-4基因(目的片段)的表达。方法:用Trizol法从上述3种肿瘤组织抽提总RNA,取出1μl作为模板进行反转录合成cDNA第一链,然后进行PCR反应。PCR产物用1.2%的琼脂糖凝胶电泳检测。结论:RT-PCR技术可以检测石蜡包埋肿瘤组织Oct-4基因。

Objective： to examine Oct-4（ goal fragment ） from the paraffin wax embedding Spermatocytoma , Dysgerminoma and Immature teratoma tissue. Methods ： we extracted total RNA from the above types of tumor tissue with Trizol, removed 1μl reverse transcriptase as a template for first strand cDNA synthesis, and then carried out PCR reaction, and checked PCR product with 1.2% agarose gel electrophoresis. Conclusion ： RT-PCR can be used to detect Oct-4 gene in paraffinembedded tumor tissue.
