摘要
目的:通过观察强力霉素对兔膝关节制动骨关节炎模型软骨Ⅱ型胶原含量和基质金属蛋白酶-13表达的影响,探讨其对骨关节炎的治疗作用及其作用机制。方法:健康成年新西兰大白兔18只,随机分为正常组、对照组和给药组,每组各6只。对照组和治疗组兔左下肢以石膏固定于伸直位4周,然后治疗组每日给予口服强力霉素治疗。4周后同时处死,观察股骨内髁关节面的病理改变,并检测软骨Ⅱ型胶原含量和基质金属蛋白酶-13水平。结果:治疗组关节软骨增生性反应较对照组明显减轻。Mankin评分和Ⅱ型胶原含量增高(P<0.01),基质金属蛋白酶-13表达减少(P<0.01)。治疗组与对照组和正常组三组之间相互比较均有显著统计学差异。结论:强力霉素可增高Ⅱ型腔原含量,改善软骨结构,其作用与强力霉素抑制软骨基质金属蛋白酶-13的表达有关。
Objective: To investigate the effect of Doxycycline on rabbit immobilized osteoarthritis and research its mechanism by determine the content of cartilage collagen - Ⅱand the expression of MMP - 13. Methods: 18 rabbits were randomly divided into normal group, control group and treatment group, with 6 rabbits in each group. Osteoarthritis model was established by immobilize the lower left limb with plaster bandage. The control group and the treatment group were immobilized for 4 weeks, then Doxycycline was orally given to treatment group once a day. All of the animals were killed after 4 weeks. Then observe the degeneration of articular cartilage of medial condyle of femur, the levels of collagen - Ⅱ and MMP -13 were tested. Results:Contrast with the control group, cartilage proliferation in the treatment group was significantly slighter. Mankin score and collagen- Ⅱ content were increased( P 〈 0.01 ), MMP- 13 was decreased (P 〈 0.01 ). There is a noticeable statistical difference among treatment group, control group and normal group. Conclusion:Doxycycline can increase the content of collagen - Ⅱ and improve the structure of cartilage, the effect relate to the inhibition of MMP - 13.
