摘要
为研究YRRM基因在国人男性中的存在状况及在男性不育病人睾丸中的表达,应用YRRM1cRNA探针,与12例无精子症病人睾丸组织mRNA进行原位杂交,结果提示YRRM基因可能参与精子生成早期减数分裂的调控。应用YRRM1和YRRM2基因的引物对10例无精子症病人和15例正常人的基因组DNA进行PCR扩增,未发现YRRM1缺失;病人及正常对照中均未发现YRRM2基因序列的存在。
By using 35 S labeled single strand RNA probe transcribed from YRRM1 cDNA clones in vitro, we perform in situ hybridization to testis tissues of 12 azoospermic adults obtained on testicular biopsies. The results suggest that the first meiotic division of early spermatocytes may be regulated by the YRRM gene. Two pairs of YRRM specific primers are applied for the PCR analysis of YRRM gene in genomic DNA of normal persons including 10 fertile males, 5 females as well as 10 azoospermic men. The results show that YRRM1 sequence only conserved in males. Even in the azoospermic patients there is no deletion of YRRM1. PCR amplification of YRRM2 shows the lack of YRRM2 in all persons involved in this study. This phenomenon has shown the ethic polymorphism of this locus where YRRM2 being deleted in all mongolians.
