浸润性宫颈癌及宫颈上皮内瘤变组织中HPV16-E7蛋白和端粒酶的表达及意义 被引量:3

摘要 目的探讨16型人乳头瘤病毒E7(HPV16-E7)蛋白和端粒酶在浸润性宫颈癌及宫颈上皮内瘤变组织中的表达以及在宫颈癌发生、发展过程中的作用。方法30例浸润性宫颈癌(ICC组);90例宫颈上皮内瘤变(CIN),其中CINⅠ(CINⅠ组)30例,CINⅡ(CINⅡ组)30例,CINⅢ(CINⅢ组)30例。采用免疫组化Elivision二步法检测各组组织标本中HPV16-E7蛋白和端粒酶的表达。另设10例正常宫颈组织标本作为对照组。结果对照组、CINⅠ组、CINⅡ组、CINⅢ组和ICC组中的HPV16-E7蛋白阳性表达率分别为30.0%、63.3%、63.3%、76.7%和96.7%,ICC组和CINⅢ组显著高于对照组,差异均有统计学意义(均P<0.05);而ICC组与CINⅢ组比较,CINⅢ组与CINⅡ组比较,差异均无统计学意义(均P>0.05)。对照组、CINⅠ组、CINⅡ组、CINⅢ组和ICC组的端粒酶阳性表达率分别为30.0%、40.0%、43.3%、70.0%和93.3%,ICC组显著高于CINⅢ组,CINⅢ显著高于CINⅡ组,差异均有统计学意义(均P<0.05);而CINⅡ组与CINⅠ组比较,CINⅠ组与对照组比较,差异均无统计学意义(均P>0.05)。浸润性宫颈癌及宫颈上皮内瘤变患者的HPV16-E7蛋白与端粒酶的表达呈正相关(r=0.376,P<0.01)。结论HPV16-E7蛋白和端粒酶的表达与宫颈癌的发生、发展有关。 Objective To explore the expression of HPV16-E7 protein and telomerase in tissue of invasive cervical carcinoma and cervical'intraepithelial neoplasia and the effect during generative,development of cervical carcinoma. Methods The expression of HPV16-E7 protein and telomerase in 30 cases of invasive cervical carcinoma(ICC group),90 cases of cervical intraepithelial neoplasia[CIN,CIN Ⅰ (CIN Ⅰ group), CIN Ⅱ (CIN Ⅱ group) and CIN Ⅲ (CIN Ⅲ group), each groups 30 cases] were detected by immunohistochemical Elivision two stages method. Another 10 cases of normal cervical tissue specimen as a control group. Results In groups of control,CIN Ⅰ group,CIN Ⅱ group, CIN Ⅲgroup and ICC group, the positive rates of HPV16-E7 protein were 30.0%,63.3%,63.3%,76.7% and 96.7% ,respectively. The positive rates of HPV16-E7 protein in ICC group and CINⅢ group were significantly higher than those in control group(P〈0.05 ,respectively). But there were no significant different in ICC group and CINⅢgroup,CIN Ⅲ group and CIN Ⅱ group(P〉0.05, respectively). And for the expression of telomerase were 30.0% ,40.0%, 43.3%,70.0% and 93.3% ,respectively. The positive rates of telomerase in ICC group were significantly higher than those in CINⅢ group(P〈0.05). And those in CINⅢ group were significantly higher than those in CIN Ⅱ group(P〈0.05) ,but there were of no significant difference in CIN Ⅱ group and CIN Ⅰ group,CIN Ⅰ group and control group(P〉0.05, respectively). The expression of HPV16-E7 protein and telomerase were positively correlated in invasive cervical carcinoma and cervical intraepithelial neoplasia(r=0. 376,P〈0. 01). Conclusion There is a close relationship between high expression of HPV16-E7 protein and telomerase during the generative, development of cervical carcinoma.
