摘要
目的评价血管紧张素转换酶(ACE)抑制剂在减轻甚至逆转高血压血管损害中的潜在作用。方法用肠系膜动脉离体灌注、HPLC和RIA检测培哚普利(一种组织渗透性ACE抑制剂)对血管合成醛固酮的影响,用RT—PCR及SouthernBlot证明培哚普利对血管表达醛固酮合成酶基因CYP11B2mRNA的作用。结果培哚普利抑制血管合成醛固酮及CYP11B2mRNA在血管中的表达。结论培哚普利治疗高血压血管损伤的机理包括抑制血管合成醛固酮。
Objective To evaluate the potential actions of angiotensin converting enzyme (ACE) inhibitors on blood vessels.Methods By means of ex vivo mesenteric artery perfusion, HPLC and RIA, aldosterone in the perfusate was measured in normal ( n =10) and perindopril (a tissue penetrative ACE inhibitor) treated ( n =10) groups. The expression of aldosterone synthase gene CYP11B2 mRNA in the vessels was determined by RT PCR and Southern blot.Results Perindopril inhibited aldosterone production and CYP11B2 mRNA expression in the blood vessels. Conclusion The inhibition of aldosterone biosynthesis is involved in the remedy of hypertensive vascular damage by perindopril.
