摘要
目的:对4种多聚物包被的球囊导管局部基因转移系统的在体转基因效果进行对比评估。方法:用由相同多聚物包被、携带不同的基因载体,及由不同的多聚物包被、携带相同的基因载体的球囊导管系统,将报告基因分别导入兔腹主动脉下段。结果:除由脂质体—腺病毒(Ad5)载体组成的基因转移系统外,其他3种系统均实现了在体基因转移。其中,由左旋多聚赖氨酸包被球囊导管—重组腺病毒载体(Ad5/LacZ)基因转移系统的基因转移效果最佳。结论:左旋多聚赖氨酸包被球囊导管—Ad5/LacZ基因转移系统是一种新型的、有效可行的在体局部基因转移系统。
Objective:This study was undertaken to compare the efficiency of in vivo gene transfer using 4 different types of polymer coated balloon catheters. Methods:The balloon catheter systems coated with the same polymer but different gene vectors,and coated with different polymers but the same gene vector were used to transfer the reporter genes into the lower part of the abdominal aorta in rabbits. Results:The reporter genes were successfully transferred into the aortic wall with all but the lipofectin coated balloon catheter.The polylysine coated balloon catheter carrying adenovirus 5(Ad5) vector system was the most effective one of the three local gene transfer systems. Conclusion:The polylysine coated balloon catheter carrying Ad5 vector is a new effective and feasible gene transfer system.
