苦瓜来源植物降糖多肽制备程序的建立及其降糖作用的实验研究 被引量:1

摘要 目的:建立提高苦瓜来源植物降糖多肽制备量的程序和方法,进一步探讨其降糖作用。方法:采用Khanna方法沉淀,经8000透析袋透析,硫酸铵盐析;上清应用CM-Sepharose柱,快速蛋白分析仪进行层析分离,洗脱峰分别采用纤维素粉薄膜层析方法进行层析分离。以所获提取物注射实验小鼠,观察其血糖变化。结果:洗脱峰经薄层层析,SDS凝胶电泳,峰1、峰3的相对分子量分别为50KD和31KD,峰2的相对分子量为11KD。注射峰2的正常小鼠和糖尿病小鼠血糖值明显下降。结论:本方法可较大程度地提高苦瓜来源植物降糖多肽的制备量,且具有良好的降糖作用。 Objective: To increase the recorvery of purification of hypoglycemic polypeptide and evaluate its hypoglycemic mechanism. Methods: salt precipitation of ammonia sulphate were used ; column chromatography of CM-Sepharose in FPLC were tested. The eluted protein peak was pooled and separated by plate chromatography of fibrocellulose. The preparation was injected into mice and blood glucose level of mice was determined. Results: the molecular weight of peak 1 is 50 000 ; which of peak 3 is 31 000 and peak 2 is 11 000. the blood level of the diabetes riceand rice injected with polypeptide of peak 2 decreased obviously. Conclusion: The methods we tested could increased the recorvery of purification of the peptide from momorcica charantia. And its effect on decrease blood glucose is obvious.
