Expression of Toll like receptor 2 in human placenta and fetal membranes

目的检测正常胎盘和胎膜组织中Toll样受体2（TLR2）的表达。方法收集5例足月剖宫分娩胎盘和胎膜样本，运用RT—PCR法检测胎盘和胎膜组织中TLR2mRNA的表达；运用免疫组织化学和免疫印迹方法检测TLR2蛋白质在胎盘和胎膜组织中的表达。结果RT-PCR显示，胎盘和胎膜组织中均有TLR2mRNA表达。免疫印迹显示TLR2在胎盘和胎膜组织中相对分子质量为50000左右的位置有一条明显的条带。免疫组织化学显示TLR2在胎盘的合体滋养细胞、胎膜的羊膜上皮细胞及平滑绒毛膜滋养细胞中有表达。结论TLR2在人胎盘和胎膜组织中有表达，提示其在妊娠期先天性免疫中可能发挥重要作用。

Objective To detect Toll like receptor 2 （TLR2） expression in tissues of normal human placenta and fetal membranes. Method Placenta and fetal membranes were collected from 5 women with intact membranes undergone elective cesarean sections at term. Expression and localization of TLR2 was detected by RT- PCR, Western blot analysis and immunohistochemistry. Results TLR2 mRNA was expressed in tissues of placenta and fetal membranes. Western blotting analysis with TLR2 antibody also yielded positive results for placenta and fetal membranes with one band site around 50 000. Immunohistochemistry showed positive TLR2 at syncytotrophoblast of placenta, cytotrophoblast of chorion and epithelial cell of amnion. Conclusion TLR2 expresses in tissues of placenta and fetal membranes. TLR2 may play an important role in the innate immunity of placenta.
