桑黄醇提物抗氧化和保护神经细胞损伤的研究 被引量:18

摘要 采用化学发光法和H2O2氧化损伤PC12细胞的模型,研究了8种桑黄子实体醇提物体外抗氧化活性及对神经细胞氧化损伤保护的作用。结果表明:8种醇提物均有良好的抗自由基活性,其中PB-10抗氧化活性最高,对超氧阴离子清除率的IC50为3.76μg/mL,对H2O2清除率的IC50为4.24μg/mL。样品在10μg/mL时对H2O2氧化损伤后的PC12细胞有较好的保护修复作用,其中PB-10活性最好,在10μg/mL时修复率为(72.5±0.3)%,150μg/mL时达到(90.7±3.6)%,且醇提物的抗氧化和氧化损伤保护作用均与样品中的黄酮含量成正相关关系。 The antioxidant activity of ethanol extracts of eight kinds of Phellinus baumii fruiting body and their protective effects against oxidative damage of nerve cells were investigated by chemilu- minometry and a model of PC12 oxidative damage by H2O2. The results showed that all the extracts had a good antioxidant activity. The extract from PB-10 had the greatest activity and its IC50 was 3.76μg/mL for superoxide anion and 4.24 μg/mL for hydrogen peroxide. All the extracts at the con- centration of 10μg/mL had good protective and repair effects on PC12 cells damaged by H202 ,and the extract from PB-10 could reach (72.5 ±0.3)% repair rate at the concentration of 10 μg/mL and up to (90.7 ± 3.6)% at 150 μg/mL. There were positive correlations between the antioxidant activities and protective effects of all the extracts and the flavone concentrations of them.
